CJC-1295 (No DAC), identical to Modified GRF (1-29), is a synthetic 29-amino-acid analog of growth-hormone-releasing hormone (GHRH). Unlike the long-acting DAC version, it lacks the albumin-binding Drug Affinity Complex, so it does not tether to serum albumin and is therefore short-acting, with a reported plasma half-life of approximately 30 minutes. CJC-1295 (No DAC) acts as an agonist at the pituitary GHRH receptor (GHRH-R), a class-B seven-transmembrane G-protein-coupled receptor on anterior-pituitary somatotrophs. Binding couples predominantly to Gs and adenylyl cyclase, raising intracellular cAMP, activating PKA, driving CREB phosphorylation and voltage-dependent calcium influx, and triggering exocytosis of stored growth hormone (GH); chronically it also supports Pit-1-mediated GH gene transcription (Mayo 1995). Downstream, pituitary GH release raises hepatic IGF-1.
