Mechanism of action
Epithalon is a synthetic tetrapeptide studied in research mainly in connection with telomere biology and cellular aging. Telomeres protect chromosome ends, and their shortening across successive cell divisions correlates in models with replicative senescence. Telomerase, including through the subunit hTERT, can lengthen telomeric DNA. In cell and tissue models, Epithalon is used to characterise the relationship between peptide signals, telomerase activity and the stability of telomere length. Laboratories that buy Epithalon receive the tetrapeptide at 10 mg per vial.
Reconstitution and handling
The lyophilizate is reconstituted in the laboratory with a suitable sterile solvent. The solvent is added slowly along the vessel wall and the preparation is mixed gently until the powder has dissolved completely. Because of the short sequence, Epithalon usually dissolves quickly. The resulting solution is processed promptly, since peptides in solution respond more readily to environmental influences. Until use, the reagent is protected from direct light. Concentration and volume are set by the laboratory in its own protocol.
In vitro research
In in vitro research, Epithalon serves as a defined reference peptide for aging and longevity studies in cell lines. Typical endpoints are telomerase activity, hTERT expression, telomere length by qPCR or TRF analysis, and senescence markers such as p16 or SA-β-Gal. Fibroblast and epithelial cell cultures are often used to compare replication limits and stress-induced aging pathways. Epithalon can be prepared in defined dilution series to test dose-dependent effects on telomere and cell-cycle parameters in controlled assays.
Quality and analytics
Each batch of Epithalon is tested for purity by HPLC, and the content is ≥99 %. An identity check confirms the tetrapeptide sequence Ala-Glu-Asp-Gly. Results are documented per batch. This analytical characterisation supports reproducible in vitro experiments when telomere and senescence parameters are compared across measurement series.
